Right now neither Kurumi nor the Yamai sisters were spirits who shared a connection with the first spirit.

Even if the Sephira crystals were taken out they would not lose their powers, with chakra in their body it would taken as the source of energy for their abilities instead of mana which is used through Sephira crystals.

Itsuki sent the ladies out as he brought in the guys he had arranged to give Issac a good time.

He had properly healed Issac and erased the pain he felt from the encounter so that he won't be mind broken or become a retard because Itsuki had done a lot to him after sending the girls back home.

But it only got worse as they slammed the second dildo down his throat, he should have died but a foreign energy in his body was constantly healing him thus he felt the constant pain.

Issac didn't know how long it had been since this nightmare had begun but he had been tortured both mentally and physically to the point that his mind had broken down.
